In the microfluidics community, poly(dimethylsiloxane) (PDMS) has become the most popular material in research laboratories because of a number of its unique properties such as optical transparency, ease to pattern with soft lithography, elastomer and relatively low price. However, in applications of biochemistry and bioanalysis, the use of PDMS can present several severe challenges. For example, microfluidic devices made from PDMS absorb small molecules from the solutions that flow through channels, which fouls the channel and complicates quantitative analysis of the solution phase; the gas permeability of PDMS can also be problematic: the evaporation of water molecules through the walls of a PDMS microchannel changes the concentration of analytes in the microchannels; and biomolecules can adsorb onto PDMS surface. In addition, most organic solvents can easily damage PDMS microchannels. Here, I will present some of our work on PDMS microchips; also I will present a novel method for the fabrication of whole-Teflon microfluidic chips which are extremely chemical-inert, anti-fouling and biocompatible.
