5. 내용 : In mammals, postnatal hematopoiesis occurs in the bone marrow (BM) and involves specialized microenvironments controlling hematopoietic stem cell (HSC) behaviour and, in particular, stem cell dormancy and self–renewal. While these processes have been linked to a number of different stromal cell types (namely osteoblasts, CXCL12-abundant reticular cells, Nestin+ mesenchymal cells, and sinusoidal endothelial cells) and signalling pathways, the composition and precise location of niches in the skeletal system remain incompletely understood. Here, we show with genetic labelling techniques, high–resolution imaging and functional experiments in mice that adult bone marrow contains specific structural and functional compartments at the periphery of the BM cavity, which we have termed hemospheres. These units are composed of endothelial, hematopoietic and mesenchymal cells, are enriched in HSCs, and allow rapid hematopoietic cell proliferation and clonal expansion. Inhibition of signalling by the receptor tyrosine kinase VEGFR2 disrupted hemospheres and reduced HSC numbers. Our results identify a previously unrecognized, vessel–associated bone marrow compartment with niche properties.
6. 약력 : Born on June 21, 1966 in Koblenz, Germany. Studies in biochemistry in Bayreuth, doctorate at the Max Planck Institute for Brain Research in Frankfurt/Main (1996), postdoctoral work at the European Molecular Biology Laboratory (EMBL) in Heidelberg. Head of an independent research group (2000) and Senior Group Leader (2005) at the Cancer Research UK London Research Institute (2000), Director at the Max Planck Institute for Molecular Biomedicine and Professor for Vascular Biology at the Westphalian Wilhelms-University since 2007.
